In response to a couple of the suggestions so far:
The Simpsons is unequivocally the greatest animation of all time. It may not be your personal favourite, but any other answer is simply wrong.
The greatest line in a film is “Your wife has a lovely neck”.
We won’t have greatest line in a song as, as @tesh has already demonstrated, YNWA has an unfair advantage on a Liverpool forum.
I’m not doing musicals, because I can’t stand them.
I’m trying to move away from films and television for the next poll; I’ll leave nominations open until 17:00 next Sunday. If nothing catches my eye by then, I’ll choose something.

Not sure if we’ve had any of these:

Greatest invention, good. Depends if you’re looking at creativity or impact. The wheel is probably up there. Internal combustion engine (which is a glorified steam engine), printing press as noted above, light bulb, radio, semiconductor, microwaves, rockets, powered flight all valid. But also all pale into insignificance next to the miraculous discovery of the glorious fusion of cheese and crackers.
Ok, the formal answer would be the The Reciprocating Engine and The Rocket Engine.
The wheel’s such a clear winner there’s no point.
